Distribution of G143A Mutations Conferring Fungicide Resistance in Virginia Populations of Parastagonospora nodorum Infecting Wheat

چکیده
Stagonospora nodorum blotch (SNB) caused by Parastagonospora is an important leaf spot disease in the mid-Atlantic United States. Disease management approaches include use of resistant varieties, cultural control, and foliar fungicides. Frequent fungicides can select for fungicide resistance within pathogen populations. Recently, first report quinone outside inhibitor (QoI) States was made based on a relatively small collection P. isolates from Virginia. The objective this study to conduct statewide, 2-year survey populations Virginia wheat quantify frequencies target-site mutation that confers QoI resistance. A total 318 were obtained collected at seven locations distributed throughout wheat-growing regions 2018 2019. previously designed pyrosequencing assay detects G143A substitution cytochrome b gene used screen presence or absence target site mutation. detected all sampled fields. Among years, ranged 5 32% (mean = 19%). Thus, QoI-resistance-conferring widespread Virginia, it occurred high frequency. Results suggest containing active ingredients may not be effective controlling SNB surrounding region, application standalone control recommended.

Fitness cost of chromosomal drug resistance-conferring mutations.
To study the cost of chromosomal drug resistance mutations to bacteria, we investigated the fitness cost of mutations that confer resistance to different classes of antibiotics affecting bacterial protein synthesis (aminocyclitols, 2-deoxystreptamines, macrolides). We used a model system based on an in vitro competition assay with defined Mycobacterium smegmatis laboratory mutants; selected mut...

Mutations conferring aminoglycoside and spectinomycin resistance in Borrelia burgdorferi.
We have isolated and characterized in vitro mutants of the Lyme disease agent Borrelia burgdorferi that are resistant to spectinomycin, kanamycin, gentamicin, or streptomycin, antibiotics that target the small subunit of the ribosome. 16S rRNA mutations A1185G and C1186U, homologous to Escherichia coli nucleotides A1191 and C1192, conferred >2,200-fold and 1,300-fold resistance to spectinomycin...
متن کاملMechanisms of streptomycin resistance: selection of mutations in the 16S rRNA gene conferring resistance.
Chromosomally acquired streptomycin resistance is frequently due to mutations in the gene encoding the ribosomal protein S12, rpsL. The presence of several rRNA operons (rrn) and a single rpsL gene in most bacterial genomes prohibits the isolation of streptomycin-resistant mutants in which resistance is mediated by mutations in the 16S rRNA gene (rrs). Three strains were constructed in this inv...

Background: Emergence Klebsiella pneumoniae resistant to quinolone antibiotics due to mutations in gyrA and parC genes created problem for treatment of patients in different hospitals in Iran. The objective of this study was to determine the amino acid substitutions of GyrA and ParC proteins in certain clonal lineages of the K. pneumoniae conferring high level quinolone resistance. Methods: One...
